What Are Anti-Gliadin Antibodies?
[dropcap]A[/dropcap]nti-gliadin antibodies (AGA) are produced by the body as an immune response against partially digested gliadin peptides (fragments) that have abnormally entered the body from the intestinal tract. Gliadin is a protein in wheat grain.
Anti-gliadin antibodies circulating in blood are measured by laboratory testing on blood that is drawn. The antibodies are both immunoglobulin A (IgA) and immunoglobulin G (IgG) types, which are termed AGA-IgA and AGA-IgG.
The presence of anti-gliadin antibodies in blood is an abnormal laboratory finding. That is, when the intestinal lining is healthy and able to keep out undigested matter from the body, there are no molecules of gliadin present to trigger the immune system for defense.
Q: Who produces anti-gliadin antibodies?
A: Persons, with or without celiac disease, can produce anti-gliadin antibodies if and when leaky gut (increased intestinal permeability) develops that then allow gliadin peptides to get into the bloodstream.
Gliadin itself causes leaky gut, but there are other causes such as various foods, intestinal infections, medications, and other diseases of the gastrointestinal tract. Whatever the cause of leaky gut, if poorly digested gliadin is present, it can slip through the leaky lining and enter the bloodstream where it can trigger an antibody response by anti-gliadin antibodies.
What Are Anti-Gliadin Antibodies In Celiac Disease and/or Gluten Sensitivity Or Non-Celiac Gluten Sensitivity?
- Relationship between elevated anti-gliadin antibodies and celiac disease. Elevated anti-gliadin antibodies (AGA) found by blood testing are a classic feature of untreated celiac disease. However, there is significant variability in the accuracy of antibodies to gliadin to signify celiac disease, whether IgA or IgG, and these tests are less specific in young children. In the reverse, some studies show antibodies to gliadin to have have a greater sensitivity for celiac disease.1
- Relationship between elevated anti-gliadin antibodies and usefulness in diagnosis of celiac disease. Because they are not normally present in the blood, their presence indicates that an immune response is being produced against gliadin, making their abnormal presence in the blood useful for identifying celiac disease in combination with the celiac anitbodies EMA (anti-endomysium antibodies) and/or tTG (anti-transglutaminase antibodies). EMA and tTG are specifically produced in celiac disease and by contrast, they are not produced in non-celiac gluten sensitivity.
- Relationship between elevated anti-gliadin antibodies and usefulness in diagnosing non-celiac gluten sensitivity. Native antigliadin antibodies IgG was positive in 66% of the children with gluten sensitivity whose biopsy results showed normal to mildly inflamed mucosa (Marsh stage 0-1).2
- Relationship between elevated anti-gliadin antibodies and leaky gut. Importantly, their presence is valuable for detecting gluten sensitivity reactions resulting from gliadin leaking into the blood as a consequence of increased intestinal permeability, whether caused by, or separate from, celiac disease.
How Prevalent Are Anti-Gliadin Antibodies In Celiac Disease and/or Gluten Sensitivity Or Non-Celiac Gluten Sensitivity?
- Celiac disease occurs in 1% of the general population.3
- IgG anti-gliadin antibodies (IgG-AGA) were positive in 56.4% of non-celiac gluten sensitivity patients and in 81.2% of celiac patients, with high antibody titers in both groups. IgA anti-gliadin antibodies (IgA-AGA) were detected in 7.7% of non-celiac gluten sensitivity patients and in 75% of celiac patients, showing lower enzyme-linked immunosorbent assay activities in non-celiac gluten sensitivity than those found in celiac disease.4

How Do Anti-Gliadin Antibodies Develop In Celiac Disease and/or Gluten Sensitivity Or Non-Celiac Gluten Sensitivity?
- Anti-gliadin antibodies are produced by the body in response to the abnormal presence of gliadin peptide molecules from wheat containing food that have leaked into the bloodstream from the small intestine and are then found in the body.
Do Anti-Gliadin Antibodies Respond To Gluten-Free Diet?
Yes. A strict gluten free diet results in disappearance of these antibodies, with or without, celiac disease.

- If you are thirsty, drink water. Add fresh, squeezed lemon to water. Lemon is anti-inflammatory, alkalizing and provides vitamin C.
- Hydration Test: Urine should be pale yellow. Fingertips should be plump, without pruning but this may not be reliable when fingers are swollen with edema. Lips should be plump, without puckering. The feeling of thirst can be unreliable.
- What is wrong with soda, coffee, tea, and alcohol? These drinks are dehydrating, increase acid, and deplete nutrients.[/box]

- Sage is also a digestive, astringent, bile stimulant and energy tonic that heals the mucosa. Drink as tea or use in cooking.
- Chamomile, lemon balm, and fennel, (as a tea) also help relieve nervous tension.
- Parsley also relieves indigestion.
- Rosemary as a tea and in cooking also is a nervous system tonic for stress and fatigue, bile stimulant, and can relieve headaches and indigestion. However, because it increases blood sugar levels, it should not be used by persons with insulin resistance or diabete.
- Thyme is also soothing remedy useful for stimulating digestion of rich, fatty foods.
Carminative Spice Remedies:
- Cloves are also antispasmodic.
- Nutmeg is also useful for indigestion.

What Do Medical Research Studies Tell About Anti-Gliadin Antibodies In Celiac Disease and/or Gluten Sensitivity Or Non-Celiac Gluten Sensitivity?
RESEARCH STUDY SUMMARIES
“Clinical, serologic, and histologic features of gluten sensitivity in children.” This study seeking to describe the clinical, serologic, and histologic characteristics of children with gluten sensitivity demonstrated findings that support the existence of gluten sensitivity in children across all ages with clinical, serologic, genetic, and histologic features similar to those of adults. Native antigliadin antibodies IgG was positive in 66% of the children with gluten sensitivity whereas, native antigliadin antibodies IgA, anti-tissue transglutaminase antibody IgA and IgG, and anti-endomysial antibody IgA were not elevated. Histology revealed normal to mildly inflamed mucosa (Marsh stage 0-1) in the children with gluten sensitivity. HLA-DQ2 was found in 7 children with gluten sensitivity.
Subjects were 15 children (10 males and 5 females; mean age, 9.6 ± 3.9 years) with gluten sensitivity who were diagnosed based on a clear-cut relationship between wheat consumption and development of symptoms, after excluding celiac disease and wheat allergy, along with 15 children with active celiac disease (5 males and 10 females; mean age, 9.1 ± 3.1 years) and 15 controls with a functional gastrointestinal disorder (6 males and 9 females; mean age, 8.6 ± 2.7 years). All children underwent celiac disease panel testing (native antigliadin antibodies IgG and IgA, anti-tissue transglutaminase antibody IgA and IgG, and anti-endomysial antibody IgA), hematologic assessment (hemoglobin, iron, ferritin, aspartate aminotransferase, erythrocyte sedimentation rate), HLA typing, and small intestinal biopsy (on a voluntary basis in the children with gluten sensitivity).
Abdominal pain was the most prevalent symptom in the children with gluten sensitivity (80%), followed by chronic diarrhea in (73%), tiredness (33%), bloating (26%), limb pain, vomiting, constipation, headache (20%), and failure to thrive (13%). No differences in nutritional, biochemical, or inflammatory markers were found between the children with gluten sensitivity and controls.2
“Serological tests in gluten sensitivity (nonceliac gluten intolerance).” This study investigated the serological pattern of gluten sensitivity in blood from 78 patients with gluten sensitivity [now called non-celiac gluten sensitivity] and 80 patients with celiac disease retrospectively assessed for immunoglobulin (Ig)G/IgA antigliadin antibodies (AGA), IgG deamidated gliadin peptide antibodies (DGP-AGA), IgA tissue transglutaminase antibodies (tTGA), and IgA endomysial antibodies (EmA). Study demonstrated that the serological pattern of non-celiac gluten sensitivity is characterized by IgG AGA positivity in more than half of cases associated to IgA AGA in a few patients, but without EmA, tTGA, and DGP-AGA, which are the specific markers of celiac disease.
IgG Anti-Gliadin Antibodies were positive in 56.4% of non-celiac gluten sensitivity patients and in 81.2% of celiac patients, with high antibody titers in both groups. IgA AGA were detected in 7.7% of non-celiac gluten sensitivity patients and in 75% of celiac patients, showing lower enzyme-linked immunosorbent assay activities in non-celiac gluten sensitivity than those found in celiac disease. Only 1 of the 78 patients with non-celiac gluten sensitivity was positive for IgG DGP-AGA (detected in 88.7% of patients with celiac disease).
IgA tTGA and IgA EmA were negative in all non-celiac gluten sensitivity patients, whereas their positivity in celiac patients was 98.7% and 95%, respectively. Patients with non-celiac gluten sensitivity displayed a variegated clinical picture with intestinal and extraintestinal symptoms (abdominal pain, bloating, diarrhea, constipation, foggy mind, tiredness, eczema/skin rash, headache, joint/muscle pain, numbness of legs/arms, depression, and anemia) together with normal or mildly abnormal small intestinal mucosa.4
“Antigliadin antibodies in the absence of celiac disease.” This study measuring antigliadin antibodies of the IgG and IgA isotypes and IgG subclasses revealed that a significantly higher proportion of the AGA-positive individuals had diarrhea and chronic fatigue and showed significantly decreased transferrin saturation, mean corpuscular volume, mean corpuscular hemaglobin, and folic acid concentrations compared with AGA-negative controls. Persistent or recurrent headaches were more common in AGA-positive patients. These findings raise the possibility that a subclinical form of gluten intolerance may be relatively common.7
“Do adults with high gliadin antibody concentrations have subclinical gluten intolerance?” Gliadin antibodies of the IgG and IgA isotypes and IgG subclasses were measured in 200 adults who were randomly selected from the Icelandic National Register. Those with the highest gliadin antibody concentrations were invited with negative controls to participate in a clinical evaluation. Neither the study subjects nor the physicians who recorded and evaluated the clinical findings were aware of the antibody levels. Significantly higher proportion of the gliadin antibody positive individuals reported unexplained attacks of diarrhoea, and IgA gliadin antibodies were associated with increased prevalence of chronic fatigue. The gliadin antibody positive group also showed significantly decreased transferrin saturation, mean corpuscular volume and mean corpuscular haemoglobin compared with the gliadin antibody negative controls. Serum folic acid concentrations were significantly lower in the IgA gliadin antibody positive individuals. On blind global assessment 15 of the 48 participants were thought to have clinical and laboratory features that are compatible with gluten sensitive enteropathy, and 14 of these were in the gliadin antibody positive group. Complaints that have not been associated with gluten intolerance had similar prevalence in both groups with the exception of persistent or recurrent headaches that were more common in the gliadin antibody positive group. These findings raise the possibility that a subclinical form of gluten intolerance may be relatively common.8
